DAL PRODUTTORE L'F-35A è la variante convenzionale di decollo e atterraggio (CTOL) ed è utilizzato principalmente dall'aeronautica degli Stati Uniti e da altre forze aeree. L'F-35A è la versione F-35 più piccola e leggera ed è l'unica variante dotata di un cannone interno. L'F-35A può eguagliare l'F-16 in termini di manovrabilità, prestazioni istantanee e ad alta gravità e superarlo in termini di furtività, autonomia con carburante interno, avionica, efficacia operativa, supporto e capacità di sopravvivenza. Ha anche un designatore laser interno e sensori a infrarossi. Nel 2013, la variante A sostituirà gli F-16 Fighting Falcons dell'USAF e nel 2028 sostituirà l'A-10 Thunderbolt II.
